If you're sure he was trained before today, take him to the vet because it sounds like something's wrong.If he gets a clean bill of health, go back to square one and potty train him like he was a new puppy. Tethered to you or crated at all times.

If he isn't neutered, he's telling you it needs to be done because he has probably begun to mark his territory. Otherwise the first answerer is right, go back to square one if he checks out with the vet.
